The test measures the amount of albumin (al-bew-min) and globulins (glah-bew-lins) in your blood. Albumin and globulins are types of protein. A small amount of your blood is put on a special plate. An electric current is sent through the plate. The protein electrophoresis test is often used to find abnormal substances called M proteins.

Serum total protein, also called plasma total protein or total protein, is a biochemical test for measuring the total amount of protein in blood plasma or serum. Protein in the plasma is made up of albumin and globulin. The globulin in turn is made up of α1, α2, β, and γ globulins. In one series, serum protein electrophoresis showed a spike or localized band in only 82 percent of patients with multiple myeloma.
